Two alternative methods for the retrieval of somatic cell populations from the mouse ovary
We present two methods for the isolation of somatic cells in the ovary. These methods are suitable for a range of applications relating to the study of folliculogenesis and mouse ovarian development.
Establishing and maintaining fertility: the importance of cell cycle arrest
(Review)
Our review focuses on the Cip/Kip family of cyclin-dependent kinase inhibitors (CKIs) in mouse gonad development. We particularly highlight recent single-cell RNA sequencing studies that show the heterogeneity of cyclin-dependent kinase inhibitors.
Primary sex determination in birds depends on DMRT1 dosage, but gonadal sex does not determine adult secondary sex characteristics
We show that DMRT1 dosage is the key sex determination factor in birds and is essential for testis development. Furthermore, we provide additional evidence that birds, in contrast to mammals, have acquired cell-autonomous sex identity (CASI) and that the sex hormone environment does not significantly influence avian secondary sexual characteristics.
The Crick COVID-19 Consortium Publications
I was part of the initial Crick COVID Consortium, established at the start of the pandemic. A number of publications have relied upon the samples we collected and processed during this time: Scalable and robust SARS-CoV-2 testing in an academic center.(Nature Biotechnology 2020), Pandemic peak SARS-CoV-2 infection and seroconversion rates in London frontline health-care workers (Lancet 2020), SARS-CoV-2 detection by a clinical diagnostic RT-LAMP assay (Wellcome Open Research 2021) and Estimating the effectiveness of routine asymptomatic PCR testing at different frequencies for the detection of SARS-CoV-2 infections (BMC Medicine 2021).
Tissue-Specific In Vivo Biotin Chromatin Immunoprecipitation with Sequencing in Zebrafish and Chicken
We describe a tissue-specific biotin ChIP-seq protocol for zebrafish and chicken embryos. We utilise AVI tagging of Transcription Factors, permitting their biotinylation by a co-expressed nuclear biotin ligase, to achieve tissue-specificity.
Genome and epigenome engineering CRISPR toolkit for in vivomodulation of cis-regulatory interactions and gene expression in the chicken embryo
The novel genome and epigenome engineering toolkit developed here enables manipulation of endogenous gene expression and enhancer activity in chicken embryos, facilitating high-resolution analysis of gene regulatory interactions in vivo.
